Non-conventional methods for galanthamine bioproduction (GALBIOPRO)

The overall aim of the project is to achieve a stable and sustained reintegration of Dr Milen Georgiev at the Host Institute (Stephan Angeloff Institute of Microbiology). This will be reached through the implementation of the scientific project. The general objective of the scientific project is to create an alternative approach for galanthamine bioproduction. This aim will be achieved through interlacing the experience of the Host Institute in the field of plant biotechnologies and the knowledge obtained from Dr Georgiev, in the field of bioprocess engineering during his previous Marie Curie project. For the achievement of scientific objectives, different strategies for improvement of galanthamine and related alkaloid volumetric yields by Leucojum aestivum differentiated organ cultures will be applied (elicitation and two phase cultivation systems will be developed); the scaling up of the biosynthetic process will be performed in different type of bioreactors (columns, temporary immersion systems); and the bioreactor culture environment will be optimised.Within the frame of the project, the possibilities for the extracts from L. aestivum to inhibit acetylcholinesterase will be checked, and the metabolomic profiling of the cultures will be performed through modern methods (GC/MS, NMR). The data obtained will lead to the identification of new compounds with pharmaceutical activity and will also contribute to the understanding of the L. aestivum primary and secondary metabolism.
